Just sharing the route awareness to my friends, who have not used this route to travel via road to Islamabad.
I covered like 11 cities in 13 days, the vehicle I used on this route was an Honda City 2012 Model. Driver duration from Karachi to Islamabad is 23 Hours, but in the period it's must to stop over every 2 hour for a small break in order to avoid continuously stressful driving.
Fuel : Every 500 meters there is a petrol pump and a cng station, so fuel isn't an issue on these roads at all. Sindh has some good restroom and washrooms, but crossing punjab has even greatest ones

My Fuel for 3700 Kms ( To and Fro ) was 240 litres of fuel ( hi-octaine and Altron regular ) - CNG was used like 11 times - Per tank was 590 rupees = 5900 rps approx ( In Sindh they use by KG and in Punjab they use by Litre , so it's the same ).
Heading from Karachi Soharab Goth, I used M-9 Motor way to Jamshoro, You can eventually use N-5 ( as GT road ) to head to Punjab, But I chose N-55 ( Indus Highway ) which is the same road that links to Rani Kot Fort, Sewan Sharif, Saan City.
Sindh Route :
There is a Diversion towards Qaim Khan city that links to N-5 Highway via Aamiri Bridge Road via the bypass towards Daulatpur - Moro - Naushahro Feroz - Kandiaro - Halani - Gadegi - Khairpur - Sukkur - Pano Aqil - Ghotki - Mirpur Matelo -Dharki - Ubaro - Chok Mari
Entering Punjab :
Ubaru ( Border of Sindh and Punjab ) - Kot Sabzal - Sanjapur - Walana Ahmed Pur - Sadiqabad - Rahim Yaar Khan - SHorkan ( M4 for 48 Kms Multan Faisalabad Motorway ) - Khanewal kabirwala road to Jhang Jhang city - Gojra - Pindi Bhittian - Lodhran - M4 - M2 Highway to ISB
Speed Limit is 110 on these roads, but better keep it under it. NHA is very active during the morning from 9 am till 11 am and then 3.30 pm to 6.30 pm.
Crossing sindh was easy as we left by 4 am and were crossed sukkur by 4 pm. We stopped over at Faiz Mahal to see the antic history about these mahal ( Faiz Mahal is a building in Khairpur, Sindh Province, Pakistan.) It was built in Khairpur Mirs in 1798 as the principal building serving as the sovereign's court for the royal palace complex of Talpur monarchs of the Khairpur dynasty. Originally it included the ruler's chambers along with 16 waiting rooms for courtiers and guest rooms for royal guests alongside the durbar and dining halls. Additionally there was the Hathi Khana for the royal elephant and the horses stables where today there is a mango orchard. This place requires a permission from D.C Khairpur ( Not a public access )
Then headed over at Bahwalpur and stayed the night there.
Having your own car has alot of benefits. For me the funny part was, that if I did not knew anything, so added a stop of a Fast food franchise and took that route to see the city at it's depth
Also Seen Khewara Salt Mine in Khewara City - This comes from Islamabad to Lahore Highway ( Motorway M2 ). Take an exit to Pid Dadan Khan Road and head to Khwara City - Follow GPS or road sign. ICI soda Ash company is also here
